Harvard and Mass General Brigham JAMA Ophthalmology Study

One important study from Mass General Brigham reported that diabetes and weight loss patients on semaglutide had higher rates of NAION. This was when compared to patients taking different medications.

For example, among overweight or obese patients, about 6.7% prescribed semaglutide developed NAION. About 0.8% using other weight loss drugs developed NAION. Denmark, Norway, and Danish Nationwide Semaglutide Studies

Scandinavian studies, including research in Denmark and Norway, have also found that people with diabetes who take semaglutide seem to have over double the risk of NAION. However, experts say the number of cases overall is still low.

GLP-1 Clinical Trial Meta-Analysis and JAMA Case Series

Other reviews also suggest a concern for the risk of optic nerve damage with some GLP-1 drugs. One 2026 analysis reportedly found that Wegovy carried the strongest signal for this problem. They aren’t automatically in the federal MDL because those cases were filed in New Jersey state courts, not in federal court. State-level Multicounty Litigation (MCL) exists to combine and manage similar lawsuits within the New Jersey state court system.
